      <title>Acute high-dose irradiation disrupts cell adhesion and Silk-Ovarioid formation in human primary ovarian cells</title>
      <description>The data repository is connected to a study that investigates the impact of X-ray treatment on primary ovarian cells (POCs) derived from cortex (cPOCs) and medulla (mPOCs) during in vitro culture. In total 50 samples collected from 5 patients were sequenced and 5 groups (i.e.,control_t1, control_t24, irradiated_t1, irradiated_t4, irradiated_t24) were included in the study.

RNA extraction was performed using the RNeasy Micro Kit (Qiagen, Germany) following the manufacturer’s instructions. Libraries were prepared using the Illumina Stranded mRNA Prep Ligation protocol (Illumina, USA) according to the manufacturer’s instruction, with 10 ng of RNA used as input. 

The dataset contains 100 fastq files from paired-end RNA sequencing, compressed with GNUzip (gzip). The dataset also contains one metadata file in xlsx format and one md5sum file in txt format. The total size of the dataset is approximately 300 GB.

Fastq files were trimmed using TrimGalore (version 0.6.1) to remove the low-quality reads before mapping. STAR aligner (version 2.7.11a) was used to map the trimmed fastq files to human genome hg38. The bam files were aligned using featureCount function in subread package (version 2.0.3) and annotated using UCSC gtf file. Processed data is deposited in GEO (GSE291604).</description>
