Data for: Single-cell sequencing reveals transcriptional and immunogenetic diversity in stereotyped chronic lymphocytic leukemia

The dataset contains raw single-cell sequencing data generated from a study of 13 individuals with chronic lymphocytic leukemia (CLL). The patients belong to three biologically defined patient subgroups in which leukemic B cells express highly similar (often near-identical) B-cell receptor immunoglobulin sequences, namely stereotyped subsets #1, #2, and #4.
The data were generated using single-cell RNA sequencing (10x Genomics Single Cell 5′ Gene Expression and V(D)J) and include information on gene expression, B-cell receptor repertoires, and T-cell receptor repertoires.
The dataset consists of raw sequencing files (FASTQ format) and associated technical metadata.
The dataset consists of 138 FASTQ files with a total size of approximately 511 GB.

Description of study design:
This is a cross-sectional study in which biological material from individuals with chronic lymphocytic leukemia was analyzed using single-cell RNA sequencing to characterize gene expression and immune receptor repertoires across three disease subgroups defined by stereotyped B-cell receptor immunoglobulin gene usage, i.e., stereotyped subsets #1, #2, and #4.

Samples were obtained from individuals diagnosed with chronic lymphocytic leukemia (CLL) belonging to stereotyped subsets #1, #2, and #4. These subsets represent biologically defined patient subgroups in which leukemic B cells express highly similar (often near-identical) B-cell receptor immunoglobulin sequences and constitute only a small proportion of CLL cases (~5%)
Due to the limited prevalence of these subsets, individuals were selected based on sample availability. Additional selection criteria included treatment-naïve status at the time of sampling and sufficient sample quality for single-cell analysis, including high cellular viability.
Single-cell suspensions were prepared from patient-derived peripheral blood mononuclear cells (PBMCs) and processed for single-cell sequencing.
